There is an unapproved moderated comment at the bottom (you probably can't see it unless you're logged in as me, but it's there). I'm having two problems:

1) I can't work out why it's in the moderation queue. As far as I can tell, discussions are permitted.

2) If I try to approve it, I get a no-permission error.

I'm probably missing some control or toggle somewhere, but I've searched and can't find it!

Even if discussions are permitted, there are separate permissions for comments. Think discussion = thread, comment = post. So there is a separate NOT-moderated permission for both discussions and comments.

Moderating wiki comments requires a separate permission from moderating wiki pages. Just make sure your user has all the "Moderator Permissions" in the wiki permissions page. If the comment still shows as moderated, and you get a no-permission error when trying to approve it, then there might be a bug. The option to approve it and the dialog to approve it should both be using the same permissions.
